2.FFPE樣本
使用芯片技術研究miRNA通常需要使用新鮮的樣本或快速冷凍的樣本。但是世界上大約有數十億份組織樣品保存在醫院或者組織樣品庫中,絕大多數是使用福爾馬林固定石蠟包埋的方法處理(Formalin-Fixed and Parrffin-Embedded,FFPE)的樣品。數量巨大的樣品蘊含著無限的科學研究的機會同時也是研究者們很棘手的研究對象。如何從這些樣品中獲取我們需要的生物信息呢? Queen’s Cancer Research Institute的研究者使用Agilent miRNA芯片對快速冷凍的樣本以及與之相匹配的相應的FFPE樣本進行了研究。 結論證實:檢測FFPE樣本中的miRNA在技術上是可行的;Agilent miRNA芯片平臺的數據顯示出其在抽提、標記和雜交過程中的高度重復性。

許多研究表明,在肝癌和癌旁正常組織之間存在miRNA表達譜的差異。另外,研究發現在人類血清血漿中存在大量穩定的miRNA。這些現象表明,大家可以尋找肝癌患者血清血漿中高含量的miRNA,以此作為生物標志物進行肝癌的早期診斷。在本研究課題中,研究人員利用miRNA芯片篩選、RT-PCR驗證等方法尋找到7個miRNA的組合,這一組合能對乙型肝癌(hepatitis B virus related HCC)患者進行早期診斷。相關成果發表在2011年12月的《Journal of Clinical Oncology》上。miRNA生物芯片技術服務由上海伯豪生物技術有限公司/生物芯片上海國家工程研究中心完成。
