2019年,Nature上再次刊登了Paul Mischel教授帶領的團隊關于eccDNA的研究成果,該研究首次提供了eccDNA的電鏡圖,近一步證實了eccDNA的環狀結構,并發現eccDNA能夠通過高度開放染色質和超遠距離調控等方式促進癌基因的轉錄。同期,Jeremy N. Rich和Peter C. Scacheri等對eccDNA中染色質結構和增強子狀態進行了分析,表明eccDNA中非編碼區的增強子功能也在eccDNA所攜帶的癌基因表達調控中發揮功能,也進一步表明了eccDNA特殊的染色質結構及其功能,這為后續的基礎與應用研究,奠定了重要的基礎。
