通過培育25個基因中攜帶前病毒整合的斑馬魚,研究人員證實,在接近一半的基因“點擊率”中,mRNA轉錄水平下降了70%以上,整合出現在外顯子或者第一個內含子中時突變的可能性最大。根據這些數據,將近1/5的整合會出現逆轉錄病毒的突變。另外,當鼠類白血病病毒特異整合到基因的第一個內含子(不與其它內含子整合)時,會出現一個強烈的誘變效應。19個基因無活性事件中的3個有胚胎缺陷。根據研究人員所夠勾勒的策略,可能F1代魚中每30個測序反應就有1起突變事件。這與傳統測序途徑(TILLING, Targeting Induced Local Lesions IN Genomes,定向誘導基因組局部突變技術)相比,鑒別斑馬魚基因突變的有效性上升20-30倍。將這種上升的有效性和F1代斑馬魚精子樣本的低溫凍藏相結合,有望得到在每個斑馬魚基因都含有突變的穩定的資源。
